    摘要:

    转化生长因子9(GDF9)与骨形成蛋白15(BMP15)通过受体介导途径,在鱼类卵细胞发育与成熟过程中发挥着重要作用。在GDF9与BMP15信号通路中,GDF9与BMP15的Ⅰ型受体基因分别为TGF-βR1a、TGF-βR1bBMPR1Ba、BMPR1Bb,两者的Ⅱ型受体基因均为BMPR2a、BMPR2b。本研究以异育银鲫为实验对象,对GDF9BMP15 的各受体基因在卵细胞中的表达模式进行研究。结果显示,在异育银鲫卵细胞发育过程中,TGF-βR1a、BMPR1BaBMPR1Bb基因mRNA水平呈先降低后升高的变化趋势,TGF-βR1b基因mRNA水平自Ⅱ期卵细胞开始显著升高,而BMPR2aBMPR2b mRNA水平则随着卵细胞的发育逐渐降低。同时,TGF-βR1b、BMPR1Bb、BMPR2aBMPR2b mRNA主要在滤泡细胞中表达,TGF-βR1aBMPR1Ba mRNA则在卵母细胞与滤泡细胞中均有表达。以上研究提示,GDF9与BMP15可通过与其受体结合,以旁分泌或自分泌参与调控硬骨鱼类卵细胞的发生、发育与成熟。

    Abstract:

    Growth differentiation factor 9(GDF9) and bone morphogenetic protein 15(BMP15) play important roles during follicular development and maturation in teleosts. In this study, we analyzed the expression pattern of type Ⅰ and Ⅱ receptors of GDF9 and BMP15 during follicle development of gibel carp. The results revealed that the expression levels of TGF-βR1a, BMPR1Ba and BMPR1Bb mRNA were first decreased and then increased during development of follicles. TGF-βR1b mRNA levels were significantly increased at the late stage of follicle development, but the expression levels of BMPR2a and BMPR2b were gradually decreased with the development of follicles. TGF-βR1b, BMPR1Bb, BMPR2a and BMPR2b mRNA were mostly expressed in follicle layer cells, but TGF-βR1a and BMPR1Ba mRNA were expressed in both oocytes and follicle layer cells. These results indicated GDF9 and BMP15 might be involved in the development of follicles by a potential paracine/autocrine signal pathway.
